    Walk-away Magnetic Bead-based PCR clean-up and Dye-Terminator Removal

This application note describes how MagSi-DNA cleanFIX is used on JANUS® Automated Workstations for high throughput nucleic acid purification. The goal was to set-up a 384-well clean-up automated solution, increasing throughput, reliability and data robustness while reducing reagent consumption and disposable costs. PerkinElmer NV and MagnaMedics Diagnostics BV created an off-the-shelf solution for automating the 384-well clean-up protocols.

Small molecules covalently coated to magnetic beads recently became very popular in several life science disciplines incl.
    • chemical proteomics
    • biomarker discovery
    • signal transduction pathway analysis
    • fishing for target proteins for target ID and/or small or large scale purification

In this application note we will outline the results from one of our clients who was interested in a generic pull out  system to analyze protein-heme interactions and heme trafficking systems within a living cell. Therefore MagnaMedics has been asked to synthesize specifically for his research heme coated magnetic beads.

MagnaMedics Diagnostics B.V. has tested its MagSi-DNA silica beads in a compatibility study using the buffers of Roche LC Nucleic Acid Isolation kits. Background of this study is the fact that clients have adopted diagnostic protocols which resulted in left over Roche buffers from these kits that.
Outcome of our study is that MagnaMedics MagSi-DNA beads are fully compatible with the original Roche beads and use of the MagSi-DNA beads with the leftover buffers will lead to significant cost savings for diagnostic clients.
